If they want to improve the function of some protein they look at single mutant effects and then determine what new effects are produced by
combining two mutants. They use the double mutatant effects to model
how multi mutant (more than double mutants) constructs might work,
produce the multimutant constructs and test them. They were able to
change CRISPR function and improve an enzymatic function 100 fold.

Intelligent design using information on single and double mutants.
